Shanghai Hengyuan Bio Report: Recently, a researcher said that some nematodes with high protein content have a longer life and live healthier than normal nematodes. These mouse models with excessive protein content appear to delay aging-related vascular dysfunction. This study is important for understanding aging and aging-related diseases.
"We found that artificially increasing or decreasing the level of KLFs (Kruppel-like transcription factor) protein family can control the lifespan of nematodes," said Nelson Hsieh MD/PHD. “Because mammals also have the same protein family, what's really exciting is that our data suggest that KLFs have similar regulatory effects on mammalian aging.â€

"Many aging-related diseases, such as high blood pressure, heart disease, and senile dementia, are associated with vascular function. We observed a decrease in KLF levels with age. Conversely, maintaining KLFs levels can prevent aging-related vascular damage," said the author, Professor Mukesh K. Jain, Associate Dean of the School of Medicine and Chief Scientific Officer of the Affiliated Hospital, added.
Further investigation, the researchers found that KLF protein works by managing autophag. Once this high quality control mechanism is lost, an aging sign will appear.
"As the cell ages, the ability to control autophagy drops," the authors explained. "This may lead to the continued accumulation of toxic proteins, which ultimately hinders cell survival. Lack of KLF protein, nematode autophagy can not maintain management, leading to premature death of nematodes."
The researchers said that the next step will explore the exact mechanisms by which autophagy in the vascular wall cells improves vascular function, and they are also looking for specific strategies for studying human KLF proteins. “As the population ages, how do the hearts and arteries that humans depend on in the later stages of life continue to work smoothly? How to design interventions that slow down the functional aging of the cardiovascular system? These are the problems we will solve in the future.â€
